IDEFIX : imaging the dynamics of fluorescence emission by two-photon excitation

FLIM, FCS and FCCS Biphotonic experiment

par Houel-Renault Ludivine - 20 juillet 2022

Thanks to a synergy of skills, the Laboratoire de Photophysique Moléculaire, the Laboratorie Charles Fabry of Institut d’Optique and the Laboratoire de Chimie Physique have developed an original device for imaging the dynamics of fluorescence emission by two-photon excitation (I.D.E.F.I.X.).

The IDEFIX microscope accessible at CPBM allows an analysis of the fluorescence emitted by the constituents of the sample or by the chromophores diffusing or present in the sample, thanks to the complementary modes of intensity imaging, lifetime (FLIM) and fluorescence cross-correlation spectroscopy (FCCS).
It is also equipped with a rheology stage.

system developed :
Excitation : Bi-photon laser Chameleon (Coherent) : tunable 690-1080nm
Detection : 2 hybrid PMA detectors (picoquant) coupled to a picoharp 300 (picoquant) : FLIM, FCS and FCCS
Rheology : RHEOPTICAD stage (CAD Instrument) : thermo-mechanical treatment
