Confocal Microscopy

TCS SP5 AOBS LEICA Microscope , equipped with UV laser and visible lasers covering the spectral range from 351 to 633 nm. It’s also coupled to a pulsed IR Ti: Sa laser for two-photon imaging.

Three photomultipliers (PMTs) are available for multi-color fluorescence intensity imaging and for spectral imaging. A FRAP license is also made available to users.

For Fluorescence Lifetime Iimaging : FLIM), the microscope is coupled to a IR Ti: Sa pulsed laser (680-1040 nm) from Coherent, and to an avalanche photodiodeconnected with a PicoHarp 3000 (PicoQuant)for time correlated single photon counting measurements (TCSPC).

It is also possible to produce FLIM spectrally resolved images thanks to a dedicated fast photomultiplier within the confocal microscope.

The PicoHarp device also allows fluorescence correlation measurements ( FCS fluorescence correlation spectroscopy ) under 1 or 2 photon laser excitation.

The confocal microscope is equipped with a thermostatically controlled and CO2-regulated enclosure which may be necessary for experiments carried out on eukaryotic cells or living bacteria.
